COMBINATION THERAPY WITH NOVEL HORMONE AGENT AND PARP INHIBITORS IN METASTATIC CASTRATION-RESISTANT PROSTATE CANCER: INITIAL EXPERIENCE AT THE UROLOGY DEPARTMENT, CHO RAY HOSPITAL

Abstract
Introduction: Metastatic castration-resistant prostate cancer (mCRPC) remains a significant clinical challenge, necessitating innovative therapeutic strategies. The combination of novel hormone agents (NHAs) and PARP inhibitors (PARPi) has been proven to have the longest survival for patients with mCRPC.
Methods: All patients with mCRPC were treated with a combination regimen including NHAs and PARPi at Cho Ray Hospital from April 2024 to April 2025. Demographic, clinical, subclinical parameters, and adverse events during follow-up time were recorded.
Results: Two patients with mCRPC treated with Abiraterone and Olaparib improved clinical status and had no progression in CT-scan imaging. The first patients have PSA decrease of more than 90% after 6 months. Moderate anemia was observed in both patients in the 3rd month of treatment. After dosage modification of Olaparib in 1-2 months, both patients recovered and continued the full dose of Olaparib.
Conclusion: This report provides the first clinical data on the use of a combination regimen including Abiraterone and Olaparib in patients with mCRPC at our institution. Initial results indicate that this combination achieves strong efficacy with notable responses in clinical and radiological measures, while adverse effects remain manageable events.
