COMPARISON OF DIFFERENT PROCESSING METHODS AND CRYOPRESERVATION SOLUTION FOR HUMAN UMBILICAL CORD TISSUE AS A SOURCE OF MESENCHYMAL STEM CELLS

Abstract
Cryopreservation of intact umbilical cord tissue is considered an optimal strategy for long-term preservation of a promising source of mesenchymal stem cells (MSCs) for regenerative medicine. This study aimed to evaluate and compare the effects of two tissue-processing sizes (1×1×1 mm and 5×2×2 mm) combined with two types of cryoprotective media (commercial medium and umbilical cord blood plasma, both supplemented with 10% DMSO) on the efficiency of umbilical cord tissue cryopreservation. Experimental results demonstrated that MSCs were successfully isolated from all cryopreserved tissue samples thawed at 1, 3, 6 months, while maintaining their biological properties (morphology, growth kinetics, immunophenotype, and differentiation potential) comparable to the fresh control group (p > 0,05). This study demonstrates the safety and efficacy of the tested cryopreservation methods for umbilical cord tissue, establishing a solid foundation for long-term storage of umbilical cord tissue in tissue banks.
