PREVALENCE AND CO-INFECTION PATTERNS OF RESPIRATORY BACTERIAL PATHOGENS IN CHILDREN USING MULTIPLEX PCR

Abstract

Background: Respiratory tract infections are common in children. Multiple bacterial pathogens may simultaneously colonize the respiratory tract, and more than one pathogen may be detected in the same specimen. However, data on the prevalence and co-detection patterns of respiratory bacterial pathogens in Vietnamese children remain limited.


Objective: To determine the prevalence and co-detection patterns of common bacterial pathogens in the respiratory tract of children using multiplex PCR.


Methods: A cross-sectional study was conducted among 2,916 children under 6 years of age at Tam Anh General Hospital. Multiplex PCR testing was performed on nasopharyngeal specimens to detect seven bacterial pathogens from January 2025 to December 2025. Clinical data and multiplex PCR results were collected and analyzed.


Results: Overall, 67.1% of specimens tested positive for at least one bacteria. S. pneumoniae (39.03%) and H. influenzae (47.43%) were the two most frequently detected bacteria. The remaining pathogens included C. pneumoniae (0.38%), L. pneumophila (0.14%), M. pneumoniae (0.1%), B. pertussis (0.07%), and B. parapertussis (0%). The highest co-detection rate was observed between S. pneumoniae and H. influenzae (19.34%), higher among children under 3 years old.


Conclusions: H. influenzae and S. pneumoniae were the most frequently detected respiratory bacteria in children under 6 years of age. Co-detection of these two pathogens was the predominant co-detection pattern, particularly among children younger than 3 years.
