Abstract

Objective: This study aimed to determine the prevalence of the BRAF V600E mutation and analyze its associations with clinical characteristics, primary tumor stage (T), and regional lymph node metastasis (N) in patients with papillary thyroid carcinoma (PTC).

Methods and materials: A retrospective study was conducted in 78 patients with PTC who had BRAF V600E testing results at Bach Mai Hospital. The mutation was detected in formalin-fixed, paraffin-embedded tissue using PCR combined with reverse molecular hybridization, while T and N stages were classified according to the 8th edition of the AJCC staging system.

Results: The mean age was 45.8 ± 14.3 years, and females accounted for 79.5% of the patients. The BRAF V600E mutation was detected in 59/78 patients (75.6%; 95% CI: 65.1–83.8%). The mutation rate did not differ significantly by age, sex, or T stage (p > 0.05). The prevalence of BRAF V600E was higher in patients with N0 disease than in those with N1 disease (85.4% vs. 64.9%), although the difference was not statistically significant by Fisher’s exact test (p = 0.063). In multivariable logistic regression, N1 was inversely associated with BRAF V600E status (adjusted OR = 0.29; 95% CI: 0.09–0.92; p = 0.035).

Conclusion: The BRAF V600E mutation was detected at a high frequency in patients with PTC and was associated with regional lymph node metastasis in the multivariable analysis.
