EVALUATION OF THE COMBINED EFFECTIVENESS OF KARYOTYPE AND CNV-SEQ IN PRENATAL DIAGNOSIS AT HAI PHONG OBSTETRICS AND GYNECOLOGY HOSPITAL

Abstract

Prenatal diagnostic testing in pregnant women at high risk of fetal chromosomal abnormalities plays a crucial role in the early detection of genetic disorders.


Objective: To evaluate the results of the combined use of karyotype and CNV-seq in prenatal diagnosis at Hai Phong Obstetrics and Gynecology Hospital.


Subjects and methods: A total of 80 high-risk pregnant women underwent amniocentesis for fetal chromosomal analysis at the Department of Laboratory Medicine, Hai Phong Obstetrics and Gynecology Hospital, from January 2024 to December 2025.


Results: The overall rate of chromosomal abnormalities was 20%, including numerical abnormalities (15%), structural abnormalities (3.7%), and combined abnormalities (1.3%). Edwards syndrome was the most common (5/16 cases), followed by Down syndrome, Klinefelter syndrome, and X chromosome abnormalities (3/16 cases). Karyotype and CNV-seq showed 100% concordance within their respective detection limits and complemented each other in identifying abnormalities beyond the scope of either method. Karyotype additionally detected one case of balanced translocation, while CNV-seq identified the origin of one marker chromosome and detected one variant of uncertain significance.


Conclusion: Combining karyotyping and CNV analysis enhances detection capability and improves the accuracy of prenatal diagnosis of chromosomal abnormalities.
