TREATMENT OUTCOME OF FIRST-LINE OSIMERTINIB IN ADVANCED STAGE NON SMALL CELL LUNG CANCER HABOURING COMMON EGFR MUTATION

Abstract

Background: Osimertinib is considered the standard first-line treatment for patients with advanced-stage, EGFR-mutated non-small cell lung cancer. However, real-world data in Vietnam are limited.


Objectives: To evaluate the outcomes of osimertinib as first-line treatment for patients with advanced EGFR-mutated non-small cell lung cancer at Tam Anh Hospital, Ho Chi Minh City.


Methods: A cross-sectional study was conducted on 44 patients diagnosed with advanced NSCLC harboring common EGFR mutations (Exon 19 deletion or L858R mutation) who received first-line Osimertinib (80mg/day) at Tam Anh Hospital, Ho Chi Minh City from 01/2022 to 08/2025.


Results: The mean age was 64 ± 10.6 years, with 65.9% being female. EGFR mutations included Exon 19 deletion (56.8%) and L858R (43.2%). The objective response rate (ORR) was 77.3% (95% CI 62.1 - 88.5); and the disease control rate (DCR) was 88.6% (95% CI: 75.4 - 96.2). The median progression-free survival (mPFS) was 14 months (95% CI: 10.91 - 17.08). Patients with baseline brain metastases achieved an mPFS of 14,8 months (95% CI: 10.3 – 19.3). Survival rates at 1, 2, and 3 years were 71.9%, 27%  16.2% respectively. Regarding toxicity, the adverse effects are mostly grade 1-2, including rash (52.3%), diarrhea (20.5%), and dry skin (43%). No treatment discontinuations due to toxicity were reported.


Conclusions: Osimertinib is an effective option and well tolerated in first-line treatment of advanced-stage non-small cell lung cancer with EGFR mutation.
